You can put anything in anything. Whether it works depends on the quality of how well the two are integrated in combination with the amount of disbelief suspension to viewer is willing to impart.

How they hold guns still rubs me wrong. But beyond that ... I don't know.
Something like Fallout: Equestria took an established world and put a crazy spin on it. It did work. You recognized the locations and the characters, and you were interested in the history because of that (I can't even say that for the real Fallout games, where I blew through holodisks like they were assigned reading). The cutie marks lets you understand a lot about a character very quickly, and provides serious issues with identity, growth, and destiny. The division of races, the little dirty secrets, it's all there already. The author of F:E also did just an exceptional job at writing the thing, with good control of pacing and character development (my only qualms were firstly, it was REALLY long, although I don't know if he could have chopped it down, given the story he was trying to tell, and secondly, something about his writing and the fact his MC is female gave me serious gender confusion). Reading through, it COULD be about a bunch of humans, as long as you accept the racial division and the monarchy and the wings and so on. But something about it being ponies gave everything a touch of extra depression and horribleness which really drives a distopian story home.

How can you get married to a copyrighted character? Twi isn't legally permitted to have opinions or desires beyond what Hasbro tells her to have. The only option I could see would be if the marriage were categorized as parody, but I don't think that would be fair to either partner. Unfortunately, I don't see any other way that marrying a copyrighted fictional pony would be considered 'fair use'.

With the exception of a few states and cities (and even those are being tied up in the court system) the only legal marriages are between a single man and a single woman. Heh, only if they're 2nd or further, there's still laws against that here. Sadly, I know a couple of marriages that are legal because of that distinction. If you added the two people's IQs together you might crack 100. And I don't even live in the southern U.S.

Of course this does bring up the question that the character Princess Luna has a darker aspect "Nightmare Moon" who was presented in the first season as the result of a god-like being, that was unappreciated and taken for granted.
